Abstract

EVARISTO, Isabel; SEABRA, Rita Costa; BAETA, José  and  PAIS, Maria Salomé. Caracterização Molecular de Proveniências de Pinus pinea L. por RAPD (Random Amplified Polymorphic DNA). Silva Lus. [online]. 2002, vol.10, n.1, pp.53-61. ISSN 0870-6352.

In 1993, "Estação Florestal Nacional" set up an experiment at Sines, with 25 stone pine provenances. Seeds from seven countries from the natural Mediterranean basin were used: Portugal, Spain, Italy, Greece, Morocco, Turkey and Israel. A total of 3750 trees were planted, 25 trees from each provenance, in six randomly repetitive blocks in an area of 0.75 ha. We selected 22 of the 25 provenances for molecular characterisation by RAPD. The RAPD profiles revealed about 51.5% of polymorphic fragments with 15 different primers. The statistical analyses revealed that twelve of the 22 provenances tested were very close, constituting a central group, probably with a common origin, not subjected to selection resulting from a breeding programme. The other ten provenances fall into three different groups, suggesting that they had been selected from the central group. The highest genetic diversity value was found in the Italian provenances, while the lowest value was found in the Portuguese provenances.

Keywords : genetic diversity; genetic distance; stone pine.
