Recent studies have shown that the substance called "Pau-ferrol A" found in young seedlings has activity against human topoisomerase II. The objective of this work was to obtain friable callus from leaf explants and nodal segments for further studies on somatic embryogenesis. The effect of different concentrations of 2,4-D and ANA alone or in combination with BAP and TDZ on callogenesis was evaluated. Our results demonstrated that both types of explants selected showed significant differences (p <0.05) in callus induction, morphology and coloring.     <p><font face = "Verdana" size = "3"><b>INTRODUÇÃO</b></font></p>

    <p><font face = "Verdana" size = "2"><i>Caesalpinia ferrea</i> Mart. (Fabaceae), popularmente conhecida como
“pau-ferro” ou “jucá”, é uma espécie arbórea amplamente utilizada na medicina popular
principalmente na região Norte e Nordeste do Brasil (Coelho-Ferreira, 2009; Oliveira
<i>et al</i>., 2010). Plântulas jovens de <i>C. ferrea </i>possuem um princípio
ativo denominado de “Pau-ferrol A” que atua contra a topoisomerase II humana, e
inibe o crescimento das células por meio da indução de apoptose, podendo ser utilizado
como importante ferramenta no tratamento da leucemia HL60 humana (Nozaki <i>et al</i>.,
2007; Ohira <i>et al</i>., 2013). Alguns grupos de pesquisas estão desenvolvendo
estudos promissores visando à obtenção de produtos fitoterápicos a serem utilizados
em seres humanos (Nakamura <i>et al</i>., 2002; Lima <i>et al</i>., 2011; Pereira
<i>et al</i>., 2012; Gallão <i>et al</i>., 2013; Lopes <i>et al</i>., 2013; Zhang
<i>et al</i>., 2013; Nawwar <i>et al</i>., 2015; Fernandes <i>et al</i>., 2016).</font></p>


    <p><font face = "Verdana" size = "2">O uso desta espécie vem aumentando de maneira
significativa nos últimos anos, o que levou à exploração indiscriminada de todos
os indivíduos adultos em idade de reprodução, além de causar a erosão genética,
reduzindo as populações nativas e comprometendo a sua regeneração natural (Benedito
<i>et al</i>., 2012). Estudos indicam que a propagação de <i>C. ferrea</i> é viável
por sementes (Scalon <i>et al</i>., 2011), por micropropagação (Silva, 2015) e por
mudas em condições <i>ex vitro</i> (Lenhard <i>et al</i>., 2013; Santos <i>et al</i>.,
2013). Entretanto, a baixa produção de sementes aliada à elevada dormência tegumentar
de <i>C. ferrea</i> (Coelho <i>et al</i>., 2010), tem limitado a produção de mudas
visando a reposição das populações naturais e os plantios <i>ex situ</i>, sendo
necessário e urgente o emprego de técnicas de reprodução, a exemplo da cultura <i>in
vitro</i>, que é uma das principais ferramentas da biotecnologia vegetal que explora
a totipotência da natureza das células vegetais (Bhojwani e Dantu, 2013a), possibilitando
a reprodução <i>in vitro </i>de genótipos superiores (George <i>et al</i>., 2008;
Loyola-Vargas e Ochoa-Alejo, 2012). Desse modo, diversas partes da planta como gemas,
meristemas apicais, embriões, segmentos de caule e raízes podem ser cultivadas <i>in
vitro</i> em meio nutritivo apropriado em ambiente asséptico (George <i>et al</i>.,
2008).</font></p>

    <p><font face = "Verdana" size = "2">A cultura de calos <i>in vitro</i>
é considerada uma técnica eficiente para obtenção de embriões somáticos, possibilitando
elevadas taxas de multiplicação que resultam em embriões que se desenvolvem em plantas
completas (Haslam e Yeung, 2011; Bhojwani e Dantu, 2013b; Isah, 2016). A organogênese
indireta de <i>C. ferrea </i>pode constituir uma alternativa
importante para multiplicação em larga escala de genótipos selecionados para desenvolvimento
de fitofármacos (Ali <i>et al.</i>, 2016).</font></p>

    <p><font face = "Verdana" size = "2">O presente
estudo teve como objetivo avaliar o efeito de diferentes concentrações das auxinas
2,4-D e ANA e sua interação com as citocininas TDZ e BAP na indução de calos, em
explantes foliares e segmentos nodais de <i>C. ferrea</i>, visando a embriogênese
somática e/ou suspensão celular.</font></p>

<body><![CDATA[<p><font face = "Verdana" size = "3"><b>MATERIAL E MÉTODOS</b></font></p>

    <p><font face = "Verdana" size = "2">Os estudos experimentais foram realizados no Laboratório
de Cultura de Tecidos Vegetais (LCTV) pertencente à Escola Superior de Tecnologia
(EST) da Universidade do Estado do Amazonas (UEA). Foram utilizados como explantes
segmentos nodais e folhas cotiledonares que foram excisados de plântulas micropropagadas
e cultivadas em meio de cultura MS (Murashige e Skoog, 1962) sem a adição de reguladores
de crescimento (Silva, 2015). Os segmentos nodais foram seccionados em fragmentos
de 1 cm e as folhas em 1 cm<sup>2</sup>, sendo que nos explantes foliares foram
feitos pequenos cortes para aumentar o contato com o regulador de crescimento.</font></p>


    <p><font face = "Verdana" size = "2">Os explantes foram inoculados em frascos de vidro
de 250 ml, contendo 40 ml de meio de cultura MS, suplementado com 30 g L<sup>-1</sup>
de sacarose, solidificado com 7 g L<sup>-1</sup> de ágar e acrescido com diferentes
concentrações das auxinas: ácido 1-naftaleno-acético (ANA) e 2,4-diclorofenoxiacético
(2,4-D), isoladamente ou em combinação com citocininas: 6-benzilaminopurina (BAP)
e tidiazuron (TDZ) (<a href = "/img/revistas/rca/v41n4/v41n4a19q1.jpg" target = "_blank">Quadro 1</a>). O pH do meio foi ajustado para 6,0 antes da autoclavagem
a 120°C por 20 minutos.</font></p>

    
<p><font face = "Verdana" size = "2">As
culturas foram mantidas no escuro por 7 dias em sala de crescimento, com temperatura
de 25°C±2°C, e então foram incubadas sob um fotoperíodo de 16 horas por 30 dias
com intensidade luminosa de 52 &#956;mol.m<sup>-2</sup>.s<sup>-1</sup>, provenientes
de duas lâmpadas fluorescentes brancas frias (GE.85W).</a> Após 30 dias de cultivo,
os explantes foram avaliados quanto à taxa de indução (%) e natureza dos calos (cor
e textura).</font></p>

    <p><font face = "Verdana" size = "2">O delineamento experimental
foi o inteiramente casualizado composto por seis repetições por tratamento, sendo
cada repetição constituída por 5 unidades experimentais (n=30). Os dados foram submetidos
à análise de variância e para a comparação das médias dos tratamentos foi aplicado
o teste de Duncan ao nível de 5% de probabilidade, utilizando o software ASSISTAT
versão 7.7.</font></p>




    <p><font face = "Verdana" size = "3"><b>RESULTADOS E DISCUSSÃO</b></font></p>

    <p><font face = "Verdana" size = "2">Os resultados (<a href = "/img/revistas/rca/v41n4/v41n4a19q1.jpg" target = "_blank">Quadro 1</a>) mostram que os explantes de <i>C.
ferrea </i>cultivados em meio MS suplementado com diferentes concentrações de 2,4-D
e ANA, isoladas ou em combinação com BAP e TDZ apresentaram diferenças significativas
(p&lt;0,05) na indução, morfologia e coloração dos calos em explantes foliares e
segmentos nodais. A iniciação de calos foi alcançada a partir dos explantes dentro
de 25-30 dias de inoculação.</font></p>

    
<p><font face = "Verdana" size = "2">A concentração de
1,0 mg L<sup>-1</sup> de 2,4-D associado com 5,0 mg L<sup>-1</sup> de TDZ no meio
MS induziu a maior porcentagem (90%) de calos friáveis em explantes  de segmentos
nodais. Enquanto que a concentração de 5,0 mg L<sup>-1</sup> de ANA em combinação
com 5,0 mg L<sup>-1</sup> de BAP  promoveu a maior porcentagem (44,67%) de calos
friáveis em explantes foliares (<a href = "/img/revistas/rca/v41n4/v41n4a19q1.jpg" target = "_blank">Quadro 1</a>). Estes resultados evidenciam que o uso
de auxinas associada à citocinina estimulam a formação de calo nos explantes de
segmentos nodais e foliares desta espécie. As respostas às concentrações de auxinas
e citocininas foram associadas à fase de desenvolvimento dos explantes, indicando
que a resposta morfogenética de um tecido <i>in vitro</i> depende da interação destas
classes de reguladores (&#346;lesak <i>et al</i>., 2017).</font></p>

    
<p><font face = "Verdana" size = "2">Alguns estudos caracterizam os calos embriogênicos pela
coloração. Aspectos translúcido-brancos ou amarelados dos calos são considerados
friáveis e com potencial para formarem embriões somáticos (Singh <i>et al</i>.,
2015). Neste estudo foram observados que os calos originados de explantes foliares
e segmentos nodais na interação de auxinas e citocininas apresentaram coloração
heterogênea, podendo identificar áreas de cores translúcidas, marrom-escuro e verdes
claros ou escuros (<a href = "/img/revistas/rca/v41n4/v41n4a19q1.jpg" target = "_blank">Quadro 1</a> e <a href = "/img/revistas/rca/v41n4/v41n4a19f1.jpg" target = "_blank">Figura 1</a>).</font></p>

    
<p><font face = "Verdana" size = "2">Essas grandes diferenças na indução de calos podem ser explicadas quando os
explantes foram mantidos nas fases escura e clara. O crescimento de tecidos vegetais
organizados <i>in vitro</i> geralmente é inibido pela luz. Entretanto, as divisões
celulares do explante e o crescimento de calo podem ser inibidos pela presença de
luz (George <i>et al</i>., 2008). No entanto, alguns explantes necessitam ser cultivados
em condições escuras, a fim de aumentar a eficiência do calo e reduzir a secreção
de compostos fenólicos, que geralmente afetam os explantes que sobrevivem (Tan <i>et
al</i>., 2010).</font></p>

<body><![CDATA[<p><font face = "Verdana" size = "2">O efeito da interação
de auxinas e citocininas associado à presença ou ausência de luz na cultura de calos
de espécies medicinais da família Fabaceae têm sido relatado na literatura. Em estudos
realizados com <i>Stryphnodendron adstringens</i> a presença de 2,4-D no meio de
cultivo favorece a indução de calos na presença e ausência de luz, porém, quando
os meios foram suplementados com baixas concentrações de BAP, na ausência de luz
produziram calos com maiores rendimentos (Castro <i>et al</i>., 2009). Werner <i>et
al.</i>(2009) observaram que os explantes de foliólolos juvenis de <i>Caesalpinia
echinata</i> cultivados com baixa concentração de 2,4-D (5 e 20 mg L<sup>-1</sup>)
e foliólolos jovens tratados com altas concentrações de 2,4-D (50 e 100 mg L<sup>-1</sup>)
geraram calos sem diferenças significativas entre luz e escuro.</font></p>

    <p><font face = "Verdana" size = "2">A formação de calos de <i>C. ferrea</i> obtidos neste estudo
é atribuída à adição exógena da combinação de reguladores de crescimento ao meio
de cultura que estimulou a proliferação celular devido os explantes de segmentos
nodais apresentarem capacidade para a indução da calogênese. Alguns autores relatam
resultados semelhantes em espécies da família Fabaceae. Fonseca <i>et al</i>. (2014)
constataram que a  interação de 10µM BAP e 2,0 µM de ANA estimularam a formação
de calos em <i>Eriythina velutina</i>. Da mesma forma, foi observado por Rady e
Nazif (1997), que a combinação de 2,0 mg L<sup>-1</sup> de 2,4-D com 2,0 mg L<sup>-1</sup>
de BAP estimulou significativamente a produção de calos obtidos de hipocótilos de
<i>Cassia acutifolia</i>. Rodrigues <i>et al.</i> (2017), em estudo realizado com
<i>Poincianella pyramidalis</i>, constataram que o uso de 1,0 mg L<sup>-1</sup> de
2,4-D associado a 2,0 mg L<sup>-1</sup> de Picloram foi eficiente na indução de
calos independentemente do tipo de explante utilizado.</font></p>



    <p><font face = "Verdana" size = "3"><b>CONCLUSÕES</b></font></p>


    <p><font face = "Verdana" size = "2">É possível promover o estabelecimento <i>in vitro
</i>de <i>Caesalpinia ferrea </i>Mart. pela formação de calos friáveis a partir
de segmentos nodais.</font></p>

    <p><font face = "Verdana" size = "2">Os explantes
foliares na ausência e presença de luz associada ao uso de reguladores de crescimento
induziram baixa porcentagem de calos friáveis.</font></p>

    <p><font face = "Verdana" size = "2"><i> </i>A formação de calos pode ser obtida em meio MS suplementado com
1,0 mg L<sup>-1</sup> 2,4-D + 5,0 mg L<sup>-1</sup> TDZ.</font></p>
